Given the size of the dust spot, it's not anywhere near the chip. Knowing the f/ratio and the pixel size of the camera, you can estimate how far the dust is from the chip. I'd check the front window of the camera, and any optics immediately ahead of the filter wheel (corrector, reducer, etc.). A can of compressed air would be the first step...
You might start with something like the Giottos Rocket Blower - hand powered, but strong enough to dislodge a lot of dust. http://giottos.com/pro_view.aspx?nId=8&TypeId=77 If you use a can of compressed air, keep the can vertical, as the propellant can be discharged onto the optical surface.
